If you listen carefully, you can easily tell the difference between Mako and Teto. For one, Mako sounds more like an instrument and not a robot. Teto sounds slightly more cutesy, while Mako sounds less nasally and has more vibrato.

It all comes with listening and getting familiar with the voices.

No, it is not. Teto was originally made as an April Fool prank. I do not know where her voice originated from; it's possible they completely synthesized it. Mako's voice was definitely human-based. It's based off of an amateur singer on Nico.

Uh, not exactly... They're freeware Vocaloids, basically. They're made from good voices, not famously good voices, in summary. Of course, there are some UTAUloids that fall into your description, too, such as Kasane Keko, whose voice bank is made from Len's and Rin's. I just wanted to make it evident that they are, originally, not related to Vocaloids at all.
