It's more or less trial and error, once you record your voice over whatever track you are singing to, play it back and listen carefully to each setting... you'll hear if it "snaps" your voice to the wrong pitch and it sounds totally off...

there is no "best auto effects" alot of good raps come from good harmony and emphasising on words, now if your talking about FX that help make your craft sound more professional, there usually should be some REVERB, Also your chorus or hook should stand out with melody, and with maybe a bit more volume.. Dont try to fix bad rapping with effects!

Just because the video is a year old doesn't mean autotune hasn't been used before then! Cher was probably the first to utilize Auto-Tune without OVER-using it. From a professional standpoint, only use AT when you need to correct MINOR errors, not every portamento slide or legato slide you sing. For this video: LESS REVERB and more chorus!

The only way to do this is in post with this effect.

The only way an effect like this has been done live effectively is with a "talk box" which was invented by Zapp & Roger in the early 80s. Snoop Dogg also used one for the recording of "Sexual Explosion".
