Using a red whip does not effect the damage. The whip's purpose is so he can move around faster. Poison's damage depends mostly on the monsters hp, which is why Grims take more damage than MDTs.

poison before explosion. if you want to, you can add 1 point to explosion so that you can finish off multiple enemies at once.

you don't train with level 1 poison. Unfortunately you are stuck with single attack skills until poison mist gets strong enough. By level 76+ you could start training with it maybe.
