Gakupo was possessed by a Demon when he killed Gakuko's mommy, as it showed in the flash back.

With each person that vows revenge, the demon waits in the host that has already fulfilled that desire(NOTE THE BLUE EYES!)

The red eyes are the mark of its next host, and it waits in their current host, like it's waiting for Gakuko to kill Gakupo, and in the end, it takes over her hot bodeh

From what I can tell, Gakupo was possessed by a demon to begin with. Notice the blue eyes.

Gakuko wanted revenge and kill him. The demon therefore take her body as a host.

The red eyes Gakuko has at 0:46 represents the hatred feelings or the desire of revenge. At 3:20, Gakupo got the same red eyes in the flashback. I believe the demon only lives in hosts who vowed revenge or something.

I'm assuming, note it's only an assumption, that it's still Gakupo's voice only altered with the gender pitch whatever part to sound like a girl. You know how Luka's voice is altered into Takoluka? Something similar to that.

Gakuko, a crossgender version of Gakupo judging by the name.

Actually, almost all vocaloids have a fanmade crossgender version of themselves which is produce by lowering or increasing by one octave or two the actual voice.

So, Kaito has Kaiko, Miku got Mikuo and Luka's one is Luki or Loki (not sure).
