I've been considering buying a Machinedrum for quite awhile now but was on the fence. This demo sold me, completely. Actually hearing what you can do with the parameter locks REALLY helps. Elektron should be paying you, man!
btw im sure you already know but when you press FUNTION mute you can cue you mutes and if you press FUNCTION mute again you can tweak some more then press function mute and BAM!

No. Reason doesn't support MIDI-out.
You could, however, use a DAW with rewire support to control reason and the machinedrum at the same time.

both samples and synthesis.... but, the sample playback goes far beyond just that- and becomes a synthesis type of its own.
