This is my idea of what could happen skipping right to the point after they meet:

Okay, so, let's just say Damon and Dean don't get along at first. Damon doesnt trust Dean because he's a hunter, Dean doesnt trust Damon and Stefan because they are vampires. Stefan tries to assure Sam and Dean that they are good vampires. Dean still doesnt trust them, but Sam starts to. Stefan and Sam bond over older, pain in the ass brothers, Damon and Dean start to like each other and everyone becomes friends.

The salvatore brothers and The Winchester brothers are so much alike...in a way. Damon and Dean are the reckless older brothers and Stefan and Sam the younger, mature brothers. If they did a crossover between these two shows, I think they could be great friends...if they got past the whole vampire and hunter thing.
