It sure is. When you are working with a pre embelished garment you want to be careful not to damage the existing print but other than that I say go for it!!
what kind of water based inks you have? 2min curing is a waste of time. water based ink should cure in seconds. 20 sec. if you just leave it and 5 sec if you're using heat gun.

Is it possible to hand-embellish an already screen-printed t-shirt (that has already been cured) with water based ink followed by heat gun curing ?

For the most part. To cure plastisol in you have to get it to 320 degrees and it will be cured.
