in yellow fever it showed sam with yellow eyes but that was just deans fears, sam is a new demon so it makes sense that he would only have dark eyes the older demons have lighter eyes, if the story line continues with sam staying as a demon i think his eyes will get lighter the more powerful he gets and all along the YED wanted sam to lead the army of hell and this episode shows that the YED started it all to raise lucifer so i think it is sams destiny to be lucifers vessel, wl see in s5!!!:)

I agree- either Lilith breaking Sam, or Sam killing Lilith will be the final seal that needs to be broken. Sam must be the meatsuit for Lucifer- that way Dean has to kill his brother to kill Lucifer etc. But i recon they're gonna leave us with a right cliff-hanger... the whole "one of the brothers is dead" has been done, so i think Dean won't kill him-it will end with Sam being full darkside... season 5 will open with them on seperate sides and Dean trying to find a way to get his brother back.
