  • @ariabm44 If you are going to use a prokaryotic plasmid in a eukaryote you need to modify it to match the linear chromosomes. That is, you will need a similar origin of replication, centromeres, and teleomeres that will allow your new artifical chromosome to replicated in step with the host cell. There's a lot of other small steps, but that's really all there is to it in the broad strokes.

    Blunt ends are just another term for DNA regions that are cut by restriction enzymes.

    If you noticed at 1:42, after the plasmids were cut, there were partial pieces of ssDNA, where the preferred gene segment attached to, on the ends? Those are considered sticky, and can be binded easily by a Ligase. Blunt ends on the other hand are very much like the name implies. There are no hanging segments, so gene fragments must bind directly to the end piece, which can prove much more difficult.
