I don't understand yet why he killed him.

Locke was killed because Ben wanted to get back to the island, and reclaim his position as leader. If Locke went back to the island, then Ben couldnt be leader.

The reason why he didnt just let Locke kill himself was because Ben had no idea how to get back. As soon as he found out that Eloise Hawking would be able to help return, he killed John.

@MadMicardo Interesting theory. But Ben told Locke that he already knew Eloise Hawking right before he killed him. And if he knew her, how could he not know about The Lamppost? He says "No" when Jack asks him if he knew about it. But then Jack turns to Eloise and asks "Is he telling the truth?" and she answers "Probably not" Also, I think Ben has realized his time as leader is up. I just think he wants to make sure Widmore doesn't find the island. Why that includes killing Locke, I have no idea.
