Personally I play it in G with normal Uke tuning ... The proper intro is impossible like this, BUT, wtf ... you can still approximate the into and the small rifs between verses is easy !!

Lyle Ritz (who played the uke on the soundtrack) played a tenor uke tuned like the top four strings of a guitar (DGBE) except the D was tuned an octave higher. Basically the same as a baritone uke with the lowest string an octave higher (re-entrant). You can play these chords on a soprano uke in standard tuning (GCEA) but it will sound higher and the chord voicings will be different. It's easier to play it in C which is a half step lower than the movie verison. D would work too.
Because Lyle Ritz plays a tenor uke tuned DGBE (with a re-entrant D) and he played the uke part on the soundtrack. The soprano that Steve Martin pretended to play in the moveie was a damaged uke that was cobbled together and used as a prop. I realize Steve Martin is a good musican, but he did not play this.
My reference is page 68 of Jim Beloff's "The Ukulele: A Visual History" which states that Ritz plays a "tenor ukulele tuned down to guitar tuning with the top string up an octave in the traditional 'my dog had fleas' tuning." Jim is good friends with Lyle and has published three books of his arrangements, so I'm sure he got this from the man himself. I've never heard of Lyle playing guitar, but he is also a bass player who played on many noted recordings including several by the Beach Boys.
last month i spent like 2 hours or more trying to figure out steve martins version! i think its cool you made this video..jus want you to know there are people out there lookin n play at that video too! i tuned down as well, much farther than half step because i would have NEVER figured out ONE string was tuned different! nice video

what makes you say it was a tenor? I think it was a Concert Uke tuned e a c# f#. Martin is seen playing a Martin soprano in the movie.

anyone paying attention? I am showing you a hot tuning, which is E A C# F#. This tuning is used on the song in the movie.
Cliff Edwards also mostly used the A tuning.
