Qantas often does this on international flights and the aircraft is designed to do this. Six engines no, but 5 is within the norm (the 5th not operating of course - its simply being ferried).

I think the success of the 787 over the disappointing sales for the A380 shows that were moving towards an aviation standard based on faster, more efficient planes with greater range rather than larger planes that airlines can pack full. I think most airlines will cap the size of the fleets with aircraft in the 777 A330 range. This is where the money and investment will be.
