It's random. You need to talk to a fisherman somewhere in the south next to a swordsman. Either you'll be playing a fish catching minigame or you'll end up fighting this boss.

That's not a very clear phrasing, but I've learned in the week since I posted that original question that what you mean to say is the boat rides are a one-way thing, not capable of going round trip.

There is a fisherman that offers you a ride between two southern regions; take the ride once, you get to fish, a second time you get the boss. Beat the boos (in 60 seconds) and you'll meet the mermaid on the next trip.

OH, THAT'S how you get to fight this boss! I had no freakin' idea! I went through the whole game with both characters, and I never once saw this boss! I felt ripped off! I had been on the boat ride only once. I definitely never knew about the mermaid. It would have been handy to go through the game with all the food that she gives you.

Still, it was false advertising. Oodako should have had at least a grab attack where he seizes Momohime, holds her for a second, then throws her or squeezes her until she can break free.
