Typically files that are in use by a program are files that are needed. And if you do boot in safe mode and delete the files, it will make programs not work properly, or the programs will just re-create them. So it's a waste of time. Only delete un-needed files that aren't in use.
the most easiest way to speed up your computer is to buy ram, delete old files, use msconfig to disable services that you don't need, or re-furbish your P.C.
Cleaning these files and cookies can improve your processors speed and performance. True, it just frees up space, but when a computer has a slower processor and low available memory every bit counts.
yea I have a 512 MB ram but I think im using too much of it on a regular bases even after a start up im using maybe half of it (im getting a 1 gig ram upgrade and atleast 200+gig HD to slave to my current)
