Counter-analogy: your body cannot deal with every invading virus the first time it encounters one - but next time around it is better equipped to fight the same virus (vaccination or immunity). Still it is probably not possible for the body to recognize new variations or mutations.
I agree with your analogy's conclusion about the impractibility of recognizing all new variations. This is why conventional AV is ineffective. I favor a whitelisting approach - only programs certified safe can run but all others are blocked The scope and volume of malware "development" (over 30K new/day) is now so high that I think it's now become a more practical and effective approach. Comodo Internet Security software and a few others have already begun implementing this approach.

i think the 3 layers are already in use in top security protects like Norton, Kaspersky, Mcafee...

Detection = Anything you want
