I inserted a custom asm block and placed it into one of my levels etc.

Then when I went to load the rom in my emulator, I selected the file which I wanted to play and it went to a black screen! How do I fix this :(

well the easiest way to make a custom boss, is to use an enemy (Custom would be nice) and use the when anything is killed level end sprite. That's one way. And other way is to use some asm hacking. That, I am not so good at tho.
