It's played several times in the game, but the first time you'll hear it is when you learn the Song of Healing from the Happy Mask Salesman when he plays it on the organ. But you won't hear it for a while becuase you're stuck as a Deku untill you complete the Swamp Temple. It's also played when oyu heal the Spirit of the Goron in his grave, the Zora washed up shore, and the man in the organ house that's been turned into a Gibdo.
