The big "circle" is shine from lighting. If you look at the manga, you'll see that the shading is darker for the pupil around the shine circle. Not completely around the bigger circle, but the bottom area, where the pupil would be located.

if it's a drawing that i do all the time, i don't use guidelines because i already know where to draw the lines. But if it's a drawing in a pose i've never drawn before or im just practicing body postures or facial expressions, then guidelines keep my drawings proportional.
