Seen Captain America. Spoilers for those who didn't watch this yet, I'm just doing some guess work here:

Near the end of the Cap movie while Howard Stark is searching for Steve, his machine stumbles across the cube in the water. Presumably he holds on to it. When he helps develop SHIELD later on, the cube likely ends up in one of their vaults. Presumably they might have researched that the cube is an Asgardian item, hence why they bring in Dr Selvig, fresh from his encounter with Thor.
