No, you don't have to solve the puzzles. For the puzzles, the question is given, then we see Layton thinking, flashes of hints of the answer, then he gets the answer (real fast) and starts solving the puzzle. Oh, and sometimes Luke tries to solve it, but in the movie he's still very new to puzzle solving, so mostly he doesn't get the answer.

Also, the person Professor Layton pointed at had a dress (or something similar [I'm not so familiar with Japanese fashion lol]) on, so it was a female Layton was pointing at.

The movie's dialogue is really fast.

Unless u have the brain of a Layton and concentrate less on the story-line and all attn. to the puzzles, then u may be able to solve it on the spot.

There are puzzles, four to be exact. It just says PUZZLE on the screen, and then Layton reads a puzzle about the mystery to them. It gives the audience enough time to solve it, and Luke, Jenis, Remi will talk about the puzzle like hints. Eventually, Layton explains the answer to them.
