I think the purpose is to give the gluing stage more surface area to ensure the book stays together. Again, this is the way I make books I don't know what would happen if you didn't follow the instructions.
book binding thread is suppsed to be used cause its waxy and less likely to snap while sewing. it is however hard to find. an awesome substitute is dental floss. seriously.
@jstluv92 it would break easily...regular thin threads wont be strong enough hold the pages together....use the thick ones...the ones you use in crocheting? i guess im right? just use the thick ones for better results.
Glue is just your run of the mill white glue/school glue/PVA. This means it might not be acid free but it doesn't bother me. When I make a book which is very precious that I want to keep forever I will do a bit more research and check out what other glue is available.

What are the tapes that you are sewing to the signatures? That is the most perplaxing thing to me. Is it a specific material or do i just use regular strips of material?

For the "tape", you could always be lazy and use black duct tape. That's what I'm doing :D

I'm assuming that the material or "tape" will allow the glue to soak in and create a stronger bond to the binding of the book.

Yes , just use fabric- its to sturdy the signatures. I'm no expert- trying making one without and see what happens.
