It is 'Aint No More Cane', The Band did a version of this song early in their career but as far as I know it's a traditional song that Levon's Dad taught him when he was young...

Especially, great toward the end (4:00) when Jerry and Janis simultaneously suggest "Alberta" (which evidently is an old folk song) as the next song to play because they're on a train through Canada and passing near Alberta. They both crack up heavily and Jerry says "Janis, I've loved ya ever since the day I saw ya." So classic.
