This lone wolf represents so much. It represents everything Mr. Fox wants to become and who he was but, at the same time, it's everything he's afraid to be. And their "fist pump" represents Mr. Fox finally connecting, letting go of all of his fears and accepting that he's not going to become what he's pictured himself as. When the wolf departs, that's Mr. Fox letting go. Letting go of his fears and what he knows that he will never become.
This scene makes me tear up everytime. It sends shivers down my spine. I think the signifigance of the scene is partially a symbol of Mr. Fox realizing that the thing he fears most is actually beautiful and he takes the time to pause and appreciate it. I also think the author of this story is trying to relay an message of conservation for all the animals. Especially the wolf who has been hated and feared and attacked for centuries. The whole movie we hear about Mr. Fox's phobia, but in the end the phobia is silly. In the end there is a common thread between all the animals, and that is to simply survive. The fist in the air is a gesture of respect and the struggle to survive and be peaceful amongst all the persecution from the humans.
