“Dippermouth Blues” AKA “Sugar foot Stomp” refers to chewing tobacco or to the person who chews tobacco. There was originally supposed to be a drum break at that point. However when they first recorded it, the drummer, Baby Dodds, missed it so the banjo player jumped in and shouted the fateful lines “Oh, play that thing.” That was the take released and every band that has played the song since has shouted the same words when they get to that part of the song. At least that's the legend.
