well in the original anime he is refered to as ed`s brother and is in that fact a he but in the manga and the l8 brotherhood anime he has no relations to the elric brothers whatsoever and also he has always had the capability to change hes appearance to that of either a man or a girl plus even thought he most often stays in the male form witch we all are most fammilliar to (even thought he looks like hes crossdressing or something...) hes real form is genderless (the big monster with 6 limbs)

The creator remade the old manga into the 03 anime because it had to many plot holes but people complained how it didnt follow the manga so he made brotherhood also. I personally prefer the 03 because you can connect to the char more and it gives meaning behind actions. Brotherhood is just more action but not as good plot.
