Two guitars in a band usually do not play the same part in a song. This sort of effect will not replace your standard rhythm/lead rock guitar combination. You're limited to whatever pattern you're playing on a single guitar with this effect.

To correctly answer your question, it just depends on the unit. I have a Digitech GNX2 floor unit and it's pitch shifting processor allows the original signal to be turned down all the way, so yeah, then you can "change the tuning" of your guitar, and hear only the new processed pitch.
the POG2 is amazing, its the same but with an extra suboctave, a few more adjustment options, smaller box (bout the same size as a big muff on its side) and a preset knob.
could this be used to "tune" your guitar, like if you were using a capo on the 1st fret or somethin, or is the original input sound played in the "background"?
