From what I can find, Killer Croc was at least in the beginning supposed to suffer from a rare condition called epidermolytic hyperkeratosis. It causes excessive growth of the skin into scale-like clumps. However, in real life it does not toughen the skin, but is instead fragile and easily harmed.
