Ok, this is a theory I have, based off of this cutscene and the next game, Demon Lord's Flute.

So, at the end of Demon Lord's Flute, Layton and *female assistant name* have saved Luke's town. Luke is so impressed that he begs his dad, Mr. Triton, to be able to go with them as assistant no. 2 (Eternal Diva). He agrees, but Luke has to return after 6 yrs. (DLF-CV=3 yrs, CV-LTT=3 yrs)

At the end of 6 yrs, Luke reluctantly returns via boat to his home town as in this cutscene.

You have really cleared things up for me. :) Maybe, they'll make a sequel to this that the father of Luke sees how happy he was with Layton that they decide to visit him, but when they do, something happens to Luke's family, so Layton and Luke have to rescue Luke's family.
