The Joker's character is supposed to be wedged somewhere between a hysterical clown and a demon, and I think Ledger's laughs, just ignoring his acting, embody what his creators would have imagined his character to be. @ 0:18 that scene shows the juxtaposition between how cruel his intentions are and yet how funny he thinks it is, and then of course the laugh he has @ 1:15 is just ridiculously demonic, nothing like that ever came out of Nicholson, Cesar Romero, or anyone.

The Joker goes much further than being just a crazy person; he ranges from a megalomaniac, to a downright cold-blooded killer who does it for nothing else than his own excitement.

In TDK he's a mass of interpretations wrapped into one, with the look he's had since Batman No. 655, and in this particular aspect I don't think The Joker could ever be outdone should another actor take on the role, and Heath Ledger worked his ass off on this part, I'll always remember his laugh - R.I.P
