well, as another commented, Ted Knight was going for a Cagney impression for The Riddler and gangsters, which is what Cagney was known to portray in movies, liked to speak quickly and often whisper through the corner of their mouths, where their bottom lip is drawn downward, so that's the reason for The Riddler's mouth drawn the way it is. I will say that the depiction of The Riddler in this series had him as a crazed man...like he wasn't all there...again, it's those facial expressions.
